NC_000023.11:g.38352554T>C was classified as Uncertain Significance for Ornithine carbamoyltransferase deficiency by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant causes a T to C nucleotide substition at the -143 position in the 5' untranslated region of the OTC gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with OTC-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Different variants occurring in the 5' untranslated region of the OTC gene have been reported to be pathogenic (PMID: 29282796). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
