Uncertain Significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by All of Us Research Program, National Institutes of Health to NM_002474.3(MYH11):c.2058+2T>C, citing ACMG Guidelines, 2015. This variant lies in the MYH11 gene (transcript NM_002474.3) at the canonical splice donor site of the intron immediately after coding-DNA position 2058, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a T to C nucleotide substitution at the +2 position of intron 17 of the MYH11 gene. Splice site prediction tools suggest that this variant may disrupt RNA splicing. To our knowledge, functional RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with MYH11-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical significance of loss-of-function MYH11 truncation and splice variants in autosomal dominant cardiovascular disorders is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
