NM_000258.3(MYL3):c.455_456insT (p.Glu152fs) was classified as Uncertain Significance for Hypertrophic cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant inserts 1 nucleotide in exon 4 of the MYL3 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with MYL3-related disorders in the literature. The role of MYL3 truncations and other loss-of-function variants in cardiovascular disorders is not yet fully understood. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
