Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000314.8(PTEN):c.731C>T (p.Pro244Leu), citing Ambry Variant Classification Scheme 2023: The p.P244L variant (also known as c.731C>T), located in coding exon 7 of the PTEN gene, results from a C to T substitution at nucleotide position 731. The proline at codon 244 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
