This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 12340, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 4114 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant changes 1 nucleotide in exon 90 of the RYR2 gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 1/248542 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The role of loss-of-function RYR2 truncation variants in cardiovascular disorders is not clearly understood. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
