NM_000368.5(TSC1):c.2626-2dup was classified as Uncertain Significance for Tuberous sclerosis syndrome by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the TSC1 gene (transcript NM_000368.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2626, duplicating one base. Submitter rationale: This variant causes a duplication of adenine at the -2 position of the splice acceptor region in intron 20 of the TSC1 gene. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with tuberous sclerosis complex (TSC) in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
