Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001128840.3(CACNA1D):c.4508T>G (p.Leu1503Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CACNA1D c.4568T>G (p.Leu1523Arg) results in a non-conservative amino acid change in the Voltage-dependent L-type calcium channel, IQ-associated domain (IPR0316490) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251440 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.4568T>G in individuals affected with Sinoatrial Node Dysfunction And Deafness and no experimental evidence demonstrating its impact on protein function have been reported.
