Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.1655A>T (p.His552Leu), citing Ambry Variant Classification Scheme 2023: The p.H552L variant (also known as c.1655A>T), located in coding exon 11 of the PMS2 gene, results from an A to T substitution at nucleotide position 1655. The histidine at codon 552 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
