Submitter rationale: Variant summary: CHEK2 c.776G>A (p.Gly259Asp) results in a non-conservative amino acid change located in the Protein kinase domain (IPR000719) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251292 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.776G>A has been reported in the literature as a VUS in settings of multigene panel testing in unaffected controls within cohorts of individuals affected with Breast/Biliary Tract Cancer (example, Momozawa_2018, Okawa_2023).
