This variant lies in the CUL7 gene (transcript NM_014780.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2398, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: CUL7 c.2398-2A>G is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 2.5e-06 in 1613964 control chromosomes (gnomAD v4.0.0). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
