This variant lies in the CYBB gene (transcript NM_000397.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 253, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: CYBB c.253-1G>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 182494 control chromosomes. To our knowledge, no occurrence of c.253-1G>C in individuals affected with X-Linked Chronic Granulomatous Disease and no experimental evidence demonstrating its impact on protein function have been reported.
