This variant lies in the CYP17A1 gene (transcript NM_000102.4) at coding-DNA position 484, where G is replaced by A; at the protein level this means replaces glycine at residue 162 with arginine — a missense variant. Submitter rationale: Variant summary: CYP17A1 c.484G>A (p.Gly162Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 250634 control chromosomes (gnomAD). To our knowledge, no occurrence of c.484G>A in individuals affected with Congenital Adrenal Hyperplasia has been reported. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant resulted in a 70% reduction in protein abundance (Capper_2018).
