Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002335.4(LRP5):c.893G>A (p.Arg298His), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the LRP5 gene (transcript NM_002335.4) at coding-DNA position 893, where G is replaced by A; at the protein level this means replaces arginine at residue 298 with histidine — a missense variant. Submitter rationale: Variant summary: LRP5 c.893G>A (p.Arg298His) results in a non-conservative amino acid change located in the EGF-like domain (IPR000742) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.893G>A has been reported in the literature in individuals affected with Vitreoretinopathy (Chen_2022). This report does not provide unequivocal conclusions about association of the variant with Familial Exudative Vitreoretinopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 36018796). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
