This variant lies in the HMBS gene (transcript NM_000190.4) at coding-DNA position 232, where A is replaced by G; at the protein level this means replaces threonine at residue 78 with alanine — a missense variant. Submitter rationale: Variant summary: HMBS c.232A>G (p.Thr78Ala) results in a non-conservative amino acid change located in the Porphobilinogen deaminase, N-terminal (IPR022417) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251494 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
