Several computational tools predict a significant impact on normal splicing: Three predict the variant creates a 5' cryptic donor site. One predict the variant strengthens that 5' cryptic donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.3e-05 in 239456 control chromosomes, including 3 heterozygotes (gnomAD v2). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.242+14C>T in individuals affected with Hearing Loss, Autosomal Dominant 72 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly benign.
