NM_000297.4(PKD2):c.2351_2354del (p.Lys784fs) was classified as Likely pathogenic for Polycystic kidney disease 2 by Molecular Genetics Department, Kulakov National Medical Research Center for Obstetrics, Gynecology and Perinatology, citing ACMG Guidelines, 2015: A previously undescribed nucleotide variant creates a frameshift p.Lys784ArgfsTer16 in the PKD2 gene. The variant was observed in heterozygous state in an individual affected with familial polycystic kidney disease. Loss-of-function variants are reported in patients with Polycystic kidney disease 2, 613095. The variant is not present in population database (gnomAD no frequency). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as likely pathogenic.
