NM_005585.5(SMAD6):c.1440dup (p.Ile481fs) was classified as Likely pathogenic for Aortic valve disease 2 by Molecular Genetics Department, Kulakov National Medical Research Center for Obstetrics, Gynecology and Perinatology, citing ACMG Guidelines, 2015: A previously undescribed nucleotide variant creates a frameshift p.Ile481HisfsTer84 in the SMAD6 gene. The variant was observed in heterozygous state in an individual affected with hypoplastic left heart syndrome. Loss-of-function variants are reported in patients with Aortic valve disease 2, 614823. The variant is not present in population database (gnomAD no frequency). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as likely pathogenic.
