Likely pathogenic for Androgen resistance syndrome — the classification assigned by Molecular Genetics Department, Kulakov National Medical Research Center for Obstetrics, Gynecology and Perinatology to NM_000044.6(AR):c.2449+1G>A, citing ACMG Guidelines, 2015. This variant lies in the AR gene (transcript NM_000044.6) at the canonical splice donor site of the intron immediately after coding-DNA position 2449, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: A previously undescribed nucleotide variant creates an alteration of the canonical splice site c.2449+1G>A in the AR gene. The variant was observed in hemizygous state in an individual affected with sex reversal. Loss-of-function hemizygous variants are reported in patients with Androgen insensitivity, 300068. The variant is not present in population database (gnomAD no frequency). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as likely pathogenic.
