Uncertain significance for Shashi-Pena syndrome — the classification assigned by Illumina Laboratory Services, Illumina to NM_018263.6(ASXL2):c.3784G>T (p.Ala1262Ser), citing ICSLVariantClassificationCriteria RUGD 01 April 2020: The ASXL2 c.3784G>T p.(Ala1262Ser) missense variant has not, to our knowledge, been reported in the peer-reviewed literature. This variant is reported in the Genome Aggregation Database in one allele at a frequency of 0.000115 in the African/African American population (version 2.1.1). The p.Ala1262 residue is located upstream of the zinc-finger domain of the protein (Shashi et al. 2016). Based on the limited evidence the c.3784G>T p.(Ala1262Ser) variant is classified as a variant of uncertain significance for Shashi-Pena syndrome.
