Likely pathogenic for TCOF1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_001371623.1(TCOF1):c.1974del (p.Val659fs): The TCOF1 c.1974delT variant is predicted to result in a frameshift and premature protein termination (p.Val659Cysfs*52). To our knowledge, this variant has not been reported in the literature or in a large population database, indicating this variant is rare. Frameshift variants in TCOF1 are expected to be pathogenic; and, a different nucleotide change resulting in a similar loss of function variant (c.1973delC, p.P658Lfsx53) has been reported in an individual with Treacher Collins syndrome (Conte et al. 2011. PubMed ID: 21951868). In summary, the c.1974del (p.Val659Cysfs*52) variant is interpreted as likely pathogenic.
