Uncertain significance for Candidiasis, familial, 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_052872.4(IL17F):c.284C>T (p.Ser95Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 95 of the IL17F protein (p.Ser95Leu). This variant is present in population databases (rs748486078, gnomAD 0.02%). This missense change has been observed in individual(s) with mucocutaneous candidiasis (PMID: 21350122). This variant is also known as p.Ser65Leu. ClinVar contains an entry for this variant (Variation ID: 30589). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects IL17F function (PMID: 21350122, 34587561). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
