Uncertain significance for GJA3-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_021954.4(GJA3):c.323_326del (p.Glu108fs): The GJA3 c.323_326delAGAG variant is predicted to result in a frameshift and premature protein termination (p.Glu108Glyfs*7). To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Frameshift variants have been documented in GJA3, but the majority occur downstream of this residue and lead to a stop loss and protein extension (Mackay et al. 1999. PubMed ID: 10205266; Li et al. 2019. PubMed ID: 31842807). Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
