This variant lies in the RAG1 gene (transcript NM_000448.3) at coding-DNA position 2825, where C is replaced by T; at the protein level this means replaces threonine at residue 942 with isoleucine — a missense variant. Submitter rationale: This sequence change replaces threonine with isoleucine at codon 942 of the RAG1 protein (p.Thr942Ile). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is present in population databases (rs762625040, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with RAG1-related conditions.
