NM_024426.6(WT1):c.887+4G>A was classified as Uncertain significance for Wilms tumor 1 by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This variant causes a G>A nucleotide substitution at the +4 position of intron 3 of the WT1 gene. This variant is also known as c.887+4G>A based on alternative transcripts, ENST00000452863 and NM_024426.6. Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been reported for this variant nor has this variant been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 7/271732 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
