This variant lies in the KCNQ1 gene (transcript NM_000218.3) at 3 bases into the intron immediately after coding-DNA position 1514, where G is replaced by A. Submitter rationale: Variant summary: KCNQ1 c.1514+3G>A alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.2e-05 in 251392 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.1514+3G>A has not been reported in the literature in individuals affected with Arrhythmia. Co-occurrence with a pathogenic variant has been reported (SCN5A c.2575C>T, p.Q859*), providing supporting evidence for a benign role. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
