Uncertain significance for ALMS1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_001378454.1(ALMS1):c.4261A>T (p.Ile1421Leu), citing ACMG Guidelines, 2015. This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 4261, where A is replaced by T; at the protein level this means replaces isoleucine at residue 1421 with leucine — a missense variant. Submitter rationale: The ALMS1 c.4264A>T variant is predicted to result in the amino acid substitution p.Thr1422Ser. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.054% of alleles in individuals of Latino descent in gnomAD, which may be too common to be a primary cause of disease. Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
