NM_000094.4(COL7A1):c.7096G>T (p.Gly2366Cys) was classified as Pathogenic for Recessive dystrophic epidermolysis bullosa by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL7A1 c.7096G>T (p.Gly2366Cys) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251362 control chromosomes (gnomAD). c.7096G>T has been observed in individuals affected with Dystrophic Epidermolysis Bullosa, Recessive (Sawamura_2005, Almaani_2011). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic/pathogenic by our lab (c.7097G>C, p.Gly2366Ala), supporting the critical relevance of codon 2366 to COL7A1 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 21448560, 37302620, 16189623). ClinVar contains an entry for this variant (Variation ID: 3036261). Based on the evidence outlined above, the variant was classified as pathogenic.
