Submitter rationale: The ABCC2 c.3G>A variant is predicted to disrupt the translation initiation site (Start Loss). To our knowledge, this variant has not been reported in the literature. However, loss-of-function variants have been reported upstream of the next putative in-frame start codon (Capalbo et al. 2019. PubMedID: 31589614; Shoda et al. 2003. PubMed ID: 14662121). This variant is reported in 0.0029% of alleles in individuals of Latino descent in gnomAD. Although we suspect that this variant might be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
