This variant lies in the GNAS gene (transcript NM_080425.4) at coding-DNA position 158, where C is replaced by G; at the protein level this means replaces proline at residue 53 with arginine — a missense variant. Submitter rationale: The GNAS c.158C>G variant is predicted to result in the amino acid substitution p.Pro53Arg. In the canonical transcript (NM_000516.7), this variant is pre-coding (c.-38304C>G). To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.025% of alleles in individuals of Latino descent in gnomAD, which may be too frequent to be an unreported primary cause of disease. Although we suspect that this variant may be benign, the clinical significance of this variant is currently classified as uncertain due to the absence of conclusive functional and genetic evidence.
