NM_004304.5(ALK):c.2494G>C (p.Asp832His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D832H variant (also known as c.2494G>C), located in coding exon 15 of the ALK gene, results from a G to C substitution at nucleotide position 2494. The aspartic acid at codon 832 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
