This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 772, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 258 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with MYBPC3-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu258*) in the MYBPC3 gene.
