Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 255 of the SLCO2A1 protein (p.Gly255Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of hypertrophic osteoarthropathy (PMID: 22197487, 28425581, 29264471). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 30187). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt SLCO2A1 protein function with a positive predictive value of 95%. This variant disrupts the p.Gly255 amino acid residue in SLCO2A1. Other variant(s) that disrupt this residue have been observed in individuals with SLCO2A1-related conditions (PMID: 22696055, 33852188), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
