Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033100.4(CDHR1):c.601G>A (p.Glu201Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 201 of the CDHR1 protein (p.Glu201Lys). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This missense change has been observed in individual(s) with inherited retinal dystrophy (PMID: 29785639, 33691693, 33946315, 35627310). ClinVar contains an entry for this variant (Variation ID: 301220). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CDHR1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
