Uncertain significance for Atrioventricular septal defect 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001308093.3(GATA4):c.842C>T (p.Thr281Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 280 of the GATA4 protein (p.Thr280Met). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with congenital heart disease (PMID: 20659440). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 30108). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GATA4 protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects GATA4 function (PMID: 23626780). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
