Pathogenic for Fanconi anemia complementation group E — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021922.3(FANCE):c.1363C>T (p.Gln455Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FANCE gene (transcript NM_021922.3) at coding-DNA position 1363, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 455 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant has not been reported in the literature in individuals affected with FANCE-related conditions. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln455*) in the FANCE gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FANCE are known to be pathogenic (PMID: 11001585, 17924555).
