NM_004304.5(ALK):c.2009C>T (p.Ser670Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S670L variant (also known as c.2009C>T), located in coding exon 11 of the ALK gene, results from a C to T substitution at nucleotide position 2009. The serine at codon 670 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
