NM_003072.5(SMARCA4):c.3181G>A (p.Glu1061Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1061K variant (also known as c.3181G>A), located in coding exon 22 of the SMARCA4 gene, results from a G to A substitution at nucleotide position 3181. The glutamic acid at codon 1061 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
