Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.7820G>A (p.Arg2607Lys), citing Ambry Variant Classification Scheme 2023: The p.R2607K variant (also known as c.7820G>A), located in coding exon 48 of the DNAH11 gene, results from a G to A substitution at nucleotide position 7820. The arginine at codon 2607 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
