Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_153614.4(DNAJB13):c.559G>A (p.Gly187Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 187 of the DNAJB13 protein (p.Gly187Ser). This variant is present in population databases (rs759748101, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with DNAJB13-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt DNAJB13 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
