Pathogenic for Pyridoxine-dependent epilepsy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001182.5(ALDH7A1):c.296G>A (p.Trp99Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with pyridoxine-dependent epilepsy (PMID: 26232297). It has also been observed to segregate with disease in related individuals. This sequence change creates a premature translational stop signal (p.Trp99*) in the ALDH7A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ALDH7A1 are known to be pathogenic (PMID: 16491085, 20554659).
