Uncertain significance for Deficiency of isobutyryl-CoA dehydrogenase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014384.3(ACAD8):c.940-13C>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ACAD8 gene (transcript NM_014384.3) at 13 bases into the intron immediately before coding-DNA position 940, where C is replaced by G. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been observed in individual(s) with isobutyryl-CoA dehydrogenase deficiency (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This sequence change falls in intron 8 of the ACAD8 gene. It does not directly change the encoded amino acid sequence of the ACAD8 protein. This variant is not present in population databases (gnomAD no frequency).
