Uncertain significance for Anemia, nonspherocytic hemolytic, due to G6PD deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001360016.2(G6PD):c.1457+37_1458-10dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the G6PD gene (transcript NM_001360016.2) at 37 bases into the intron immediately after coding-DNA position 1457 through 10 bases into the intron immediately before coding-DNA position 1458, duplicating this region. Submitter rationale: This sequence change falls in intron 12 of the G6PD gene. It does not directly change the encoded amino acid sequence of the G6PD protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with G6PD-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
