Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006579.3(EBP):c.194C>T (p.Ser65Phe), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs782294297, gnomAD 0.005%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with EBP-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt EBP protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 65 of the EBP protein (p.Ser65Phe).
