NM_004304.5(ALK):c.609G>C (p.Arg203Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R203S variant (also known as c.609G>C), located in coding exon 1 of the ALK gene, results from a G to C substitution at nucleotide position 609. The arginine at codon 203 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
