Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002055.5(GFAP):c.1171+463C>T, citing Invitae Variant Classification Sherloc (09022015): The GFAP gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_001131019.2, and corresponds to NM_002055.4:c.1171+463C>T in the primary transcript. This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 427 of the GFAP protein (p.Pro427Leu). This variant is present in population databases (rs765781978, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with GFAP-related conditions. ClinVar contains an entry for this variant (Variation ID: 2984585). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
