Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001039876.3(SYNE4):c.662_666del (p.Glu221fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu221Glyfs*18) in the SYNE4 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SYNE4 are known to be pathogenic (PMID: 23348741, 28958982). This variant is present in population databases (rs772404105, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with SYNE4-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
