This variant lies in the DPYD gene (transcript NM_000110.4) at 11 bases into the intron immediately before coding-DNA position 2180, where G is replaced by A. Submitter rationale: Variant summary: DPYD c.2180-11G>A alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00011 in 250966 control chromosomes, predominantly at a frequency of 0.0016 within the African or African-American subpopulation in the gnomAD database. This frequency is not significantly higher than expected for a pathogenic variant in DPYD causing Dihydropyrimidine Dehydrogenase Deficiency (0.0016 vs 0.0025), allowing no conclusion about variant significance.
