Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042492.3(NF1):c.6236del (p.Ala2079fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 6236, deleting one base; at the protein level this means shifts the reading frame starting at alanine residue 2079, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with NF1-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change creates a premature translational stop signal (p.Ala2058Valfs*3) in the NF1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538).
