Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000202.8(IDS):c.31C>T (p.Leu11Phe), citing Ambry Variant Classification Scheme 2023: The p.L11F variant (also known as c.31C>T), located in coding exon 1 of the IDS gene, results from a C to T substitution at nucleotide position 31. The leucine at codon 11 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
