This variant lies in the LEPR gene (transcript NM_002303.6) at coding-DNA position 536, where G is replaced by A; at the protein level this means replaces glycine at residue 179 with aspartic acid — a missense variant. Submitter rationale: The LEPR c.536G>A variant is predicted to result in the amino acid substitution p.Gly179Asp. This variant was observed in a cohort of obese individuals, and in vitro functional studies showed inconclusive evidence of loss of function (Supplemental Data Set, Shah et al. 2023.
